How to Reconstitute AICAR: Step-by-Step Laboratory Preparation Guide
AICAR (5-Aminoimidazole-4-carboxamide ribonucleoside, Acadesine, MW 258.23 g/mol, CAS 2627-69-2) is a cell-permeable AMP analogue and direct AMPK activator used extensively in metabolic pathway and cell signalling research. It is supplied as a lyophilised powder in a 10mL sterile glass vial containing 50mg. AICAR differs from peptide research compounds in that it is a nucleoside analogue rather than a peptide — it is exceptionally water-soluble and requires no special reconstitution technique.
Materials Required
- AICAR 50mg lyophilised vial
- Sterile water or PBS for reconstitution
- 1mL or 2mL sterile syringe with needle
- Alcohol swabs (70% isopropyl)
- Nitrile gloves
- Permanent marker for labelling
- Refrigerator at 2–8°C for storage
Important Note on Solvent Selection
AICAR is a small nucleoside analogue with exceptional aqueous solubility (>100mg/mL in water). It can be reconstituted in sterile water, PBS, or BAC water — sterile water or PBS is commonly preferred as AICAR does not require the preservative function of BAC water for single-experiment usage. For stock solutions intended for multi-week use, BAC water is acceptable. AICAR does not require DMSO or acetic acid co-solvents.
Step-by-Step Reconstitution Protocol
Step 1 — Prepare workspace. Wipe work surface with 70% isopropyl alcohol. Allow to dry. Wear nitrile gloves.
Step 2 — Swab vial stopper. Clean the rubber stopper with a fresh alcohol swab. Air dry 10 seconds.
Step 3 — Draw 2mL of sterile water or PBS. For a 50mg vial, 2mL yields a 25mg/mL stock — a practical high-concentration stock for serial dilution to millimolar working concentrations used in AMPK activation studies.
Step 4 — Inject along the vial wall. Direct the solvent slowly down the inner glass wall over 10 seconds.
Step 5 — Dissolve. AICAR dissolves instantly due to its high aqueous solubility. Swirl briefly for 15 seconds. The solution should be clear and colourless immediately. Do not shake.
Step 6 — Label. Record reconstitution date, concentration, solvent, and initials.
Resulting Concentration
Adding 2mL sterile water to a 50mg AICAR vial yields a stock concentration of 25mg/mL. For AMPK activation assays in C2C12, HepG2, or 3T3-L1 cell model preparations, working concentrations of 0.1mM–2mM are standard, prepared by serial dilution into cell culture media. Note that AICAR working concentrations for cell models are typically in the millimolar range — significantly higher than peptide research compounds which operate in the nanomolar range.
Concentration Reference Table
| Water/PBS Volume Added | Resulting Concentration |
|---|---|
| 1mL | 50mg/mL (~194mM) |
| 2mL | 25mg/mL (~97mM) |
| 5mL | 10mg/mL (~39mM) |
Storage Conditions and Stability
Store lyophilised AICAR at −20°C prior to reconstitution, protected from moisture. Once reconstituted in sterile water or PBS, store at 2–8°C. AICAR in aqueous solution is stable for up to 28 days under refrigeration. Unlike peptide compounds, AICAR's nucleoside structure is stable across a wide pH range (3.0–9.0) and does not require carrier protein supplementation. Protect from repeated freeze-thaw cycles by preparing single-use aliquots for −80°C storage.
Research Application Notes
AICAR's mechanism of action as an AMPK activator depends on intracellular phosphorylation to ZMP (AICA ribonucleotide monophosphate) by adenosine kinase — this phosphorylation step occurs inside the cell, so AICAR must be cell-permeable and present in the media at sufficient concentration to produce intracellular ZMP accumulation that mimics AMP signalling. Standard AMPK activation in C2C12 and hepatocyte cell models uses 0.5–1mM AICAR for 30–120 minute incubation periods, confirmed by AMPK Thr172 phosphorylation immunoblot. Include compound C (dorsomorphin, 10µM) as a selective AMPK inhibitor control to confirm AMPK pathway dependence of observed cellular responses. AICAR 50mg vials are available from Pinnacle Peptides, verified at ≥99% purity by HPLC and mass spectrometry.
